MOTS-c (Mitochondrial Open Reading Frame of the Twelve S rRNA Type-c) is a mitochondrial-derived peptide (MDP) consisting of 16 amino acids. It was first identified and characterised in 2015 by researchers at the University of Southern California as a novel signalling peptide encoded within the mitochondrial genome — specifically the 12S rRNA gene. MOTS-c is one of a small number of peptides known to be encoded by mitochondrial DNA rather than nuclear DNA.
Published peer-reviewed research has investigated MOTS-c’s role in cellular energy homeostasis and metabolic regulation. Studies have examined its activation of the AMPK (5′ AMP-activated protein kinase) signalling pathway, a central regulator of cellular energy balance. Research has explored its effects on folate-methionine cycle metabolites, de novo purine synthesis, and AICAR accumulation in cellular models. Additional published studies in journals including Cell Metabolism have investigated its role in skeletal muscle metabolism, exercise response, insulin sensitivity, and age-related metabolic decline in animal models. MOTS-c has been described in published literature as an “exercise mimetic” due to its observed effects on metabolic pathways typically activated during physical activity.
